Optimal production of sulphuric acid by Thiobacillus thiooxidans using response surface methodology

چکیده
Response surface methodology was applied to determine the optimal medium composition for sulphuric acid production by an indigenous Thiobacillus thiooxidans. With a view to reducing the number of experiments and obtaining the mutual interactions between the variables, a 24 full-factorial central composite design was employed for experimental design and analysis of the results. Four independent variables, KH2PO4, (NH4)2SO4, MgSO4 and elemental sulphur, in the growth medium were tested. Elemental sulphur was found to exhibit positive correlation with the sulphuric acid yield, whereas the other three variables showed no significant effect. No mutual interactions were found between elemental sulphur and each of the other three variables. The optimal composition of the growth medium to achieve the optimal production of sulphuric acid was determined as follows (g/l): KH2PO4 = 3.5, (NH4)2SO4 = 4.9, MgSO4 = 0.7 and elemental sulphur = 23.7. The corresponding sulphuric acid production and dry cell weight were 38 662 ppm and 0.7 g/l, which are about 2.6and 1.75-fold increase, respectively, compared with those using the conventional T. thiooxidans optimum growth medium. © 2003 Elsevier Ltd. Dose–response assessment of metal toxicity upon indigenous Thiobacillus thiooxidans BC1
This study provides a first attempt from a toxicological perspective to put forward, in general terms and explanations, the toxicity series of Cd(II), Cu(II) and Zn(II) to Thiobacillus thiooxidans BC1. Sulphur oxidation and sulphuric acid production are strongly related to microbial growth at pH less than 4. Dose–response analysis on chronic and acute toxicity (e.g. EC20, median effective dose ...
متن کاملA simple medium modification for isolation, growth and enumeration of Acidithiobacillus thiooxidans (syn. Thiobacillus thiooxidans) from water samples.
High concentrations of H(2)S in groundwater are commonly removed using Biological Trickling Filter (BTF) that contains high numbers of biofilm immobilized sulfur oxidizing bacteria (mainly Thiobacillus thiooxidans). BTF performance requires continuous monitoring of these bacteria at several sampling points. The Most Probable Number (MPN) technique is at the moment the method of choice to enumer...
متن کاملEffect of pH on sulfite oxidation by Thiobacillus thiooxidans cells with sulfurous acid or sulfur dioxide as a possible substrate.
The oxidation of sulfite by Thiobacillus thiooxidans was studied at various pH values with changing concentrations of potassium sulfite. The optimal pH for sulfite oxidation by cells was a function of sulfite concentrations, rising with increasing substrate concentrations, while that by the cell extracts was unaffected. The sulfite oxidation by cells was inhibited at high sulfite concentrations...
متن کاملDegradation of oil shale by sulfur-oxidizing bacteria.
Approximately 40% of oil shale can be solubilized by the action of sulfur-oxidizing bacteria. Thiobacillus thiooxidans and Thiobacillus concretivorous are equally effective in solubilization. Continuous leaching experiments show that this process can be completed within 14 days. The growth of Thiobacillus and the production of acid were measured under several conditions. Almost all of the CaMg(...
